H-Gly-Gly-His-Ala-OH is a peptide of four amino acids. It has been shown experimentally that the side chain of His is always oriented in the same direction, regardless of the conformation. The proton shift constants and vicinal coupling constants for Gly, Gly and Ala are constant, while His has a variable proton shift constant. The experimental parameters for the molecule have been rationalized by using conformational shifts to explain their variability. For example, the proton shift constants have been found to be dependent on the rotation about an axis perpendicular to the peptide backbone. H-Ala-Gly-Gly-His-OH is a pentapeptide with five amino acids that differs from HGGHAOH by having Ala instead of His at position 1. The vicinal coupling constants are different and so are other experimental parameters.
