Leflunomide è un farmaco immunomodulatore utilizzato principalmente nel trattamento dell'artrite reumatoide e dell'artrite psoriasica. Funziona come un pro-farmaco, il che significa che viene metabolizzato nel corpo nella sua forma attiva, A77 1726, che inibisce l'enzima diidroorotato deidrogenasi. Questa inibizione interrompe la sintesi de novo delle pirimidine, portando a una riduzione della proliferazione dei linfociti e alla modulazione delle risposte immunitarie.
