Boc-N-methyl-D-leucine is a macrocyclic peptide that has been shown to bind to the active site of a protein called D-alanyl-D-alanine ligase. The binding of this drug to the enzyme prevents the formation of an ATP molecule, which is required for the catalysis of peptide bond formation. Boc-N-methyl-D-leucine also mimics vancomycin, which is an antibiotic that binds to D-alanyl-D-alanine ligase and inhibits its activity. This drug has been shown to inhibit bacterial growth in vitro and in vivo at micromolar concentrations.
