Fmoc-L-cyclohexylglycine is a potent inhibitor of the NS3 protease. It constitutes one of the first anti-hepatitis C virus (HCV) inhibitors that are potent and selective. The drug has been shown to inhibit HCV replication in vitro with low nanomolar potency and high selectivity. Fmoc-L-cyclohexylglycine is a trisubstituted cyclopentane, which binds to an active site on the NS3 protease and blocks the enzyme's catalytic activity. Fmoc-L-cyclohexylglycine also inhibits other proteases, such as HIV protease and influenza virus NS1 proteinase, by binding to a template region within their active sites.
