Phorbol-12-myristate-13-acetate is a phorbol ester that inhibits the activity of protein kinase C. It binds to the response element on DNA and blocks RNA synthesis, leading to cell death. Phorbol 12-myristate 13-acetate has been shown to inhibit the proliferation of human squamous cells in culture, which may be caused by its ability to inhibit the cell cycle. This drug also has been shown to inhibit the production of cytokines such as interleukin-1β and tumor necrosis factor α (TNFα) in human polymorphonuclear leukocytes (PMNLs).
