Prototribestin is a saponin that has been shown to have anti-inflammatory properties. Prototribestin was isolated from the plant Tribulus terrestris, which grows in India. Prototribestin has been shown to inhibit inflammatory enzymes such as phospholipase A2, cyclooxygenase (COX-1 and COX-2), lipoxygenase, and nitric oxide synthase. Prototribestin also inhibits the production of proinflammatory prostaglandins and leukotrienes, while increasing the production of anti-inflammatory prostaglandins. Prototribestin also inhibits TNF-α and IL-1β induced expression of ICAM-1 and VCAM-1 in endothelial cells.
