(1R,4R,9R,10R)-2,8,16-Trihydroxy-5,5,9-trimethyl-14-methylidenetetracyclo[11.2.1.01,10.04,9]hexadecan-15-one is a natural product that is found in the roots of the plant Isodon rugulosus and has been shown to have anti-inflammatory activities in vitro. This compound binds to the p38 mitogen activated protein kinase (MAPK) pathway and inhibits its activation by reducing phosphorylation of the p38 MAPK molecule. It also inhibits the production of proinflammatory cytokines by inhibiting nuclear factor kappa B (NFκB) translocation. The mechanism of action of this compound may be due to its ability to inhibit NFκB translocation and subsequent transcriptional activity or its ability to inhibit IL-1β production by blocking NFκB signaling pathways.
