Description:

GS-9256 is a potent and selective protein inhibitor that blocks the NS3 protease of hepatitis C virus (HCV) and may be used in the treatment of chronic HCV infection. GS-9256 has a macrocyclic structure and binds to the active site of the NS3 protease, which is required for viral replication. The drug also inhibits other serine proteases such as thrombin, trypsin, factor VIIIa, and plasmin. GS-9256 has been shown to be safe and well tolerated in clinical trials with no significant adverse effects or serious side effects observed. It does not induce any resistance or cross-resistance to other antiviral drugs used for HCV treatment such as pegylated interferon alpha-2b or ribavirin. GS-9256 is rapidly absorbed from the gastrointestinal tract into circulation with excellent oral bioavailability. It is metabolized by CYP3A4/5 and eliminated by renal excretion.
