S-(5²-adenosyl)-L-methionine iodide (SAMMI) is a cytosolic metabolite of the amino acid methionine. It has been found to be immunoreactive in human lung tissue, and its presence is correlated with taurocholate concentration. SAMMI inhibits catechol-O-methyltransferase (COMT), which leads to increased levels of noradrenaline and other catecholamines that are involved in the pathogenesis of lung cancer. SAMMI binds to human lung at the site of injury, leading to ligation of DNA and inhibition of protein synthesis. The effect on protein synthesis is mediated by its ability to repress transcriptional activity through inhibition of RNA polymerase II binding to the promoter regions for genes important in cell proliferation.
