2-Amino-1-methylbenzimidazole (2AMB) is an antibacterial drug that inhibits bacterial growth by binding to the 50S ribosomal subunit. It has been shown to have a structure similar to the antibiotic chloramphenicol, which binds at the same site as 2AMB. A crystallographic study of 2AMB has shown it forms a complex with copper ions, and this may be the reason for its antimicrobial activity.
2AMB is not toxic to human cells in culture and can inhibit the growth of cancer cells. However, it is toxic to mice, leading to death after oral administration. The toxicity of 2AMB in mice may be due to its ability to induce apoptosis and DNA damage in peripheral blood lymphocytes. This drug also inhibits bacterial cell division by inhibiting RNA synthesis and protein synthesis.
