Bz-Thr-OMe is a peptidyl prodrug which is hydrolyzed to form the corresponding carboxylic acid group, amide, and ionizable carboxylate. The peptidyl prodrug has a transpeptidation reaction with the amino acid residue of the target protein. Bz-Thr-OMe also reacts at an acidic ph with a carboxylic acid group of its substrate. This reaction forms an imine intermediate which is converted to the corresponding amide by hydrolysis. The rate of this reaction is dependent on the concentration of hydrophobic groups in the vicinity of the reactive site.
