Calcineurin is a cytosolic protein that regulates the production of cytokines and other inflammatory mediators. It is an enzyme, which is activated by phosphorylation and inactivated by dephosphorylation. Calcineurin substrates are used to study calcineurin activity in biological systems. Activated calcineurin promotes the expression of genes involved in axonal growth and mitochondrial membrane potential, and inhibits the transcriptional regulation of genes encoding proteins required for renal filtration. One such substrate, p-nitrophenyl phosphate (pNPP), is phosphorylated to become active, while another substrate, cycloheximide (CHX), prevents protein synthesis by blocking the ribosome at the start codon and inhibiting translation elongation.
