Description

Pemetrexed is a drug that belongs to the class of drugs called antifolates. It is used in chemotherapy to treat cancer, especially in combination with other medicines. Pemetrexed inhibits cell growth by interfering with the synthesis of DNA and RNA. It binds to the enzyme dihydrofolate reductase, which is involved in the production of one-carbon units needed for DNA synthesis, and inhibits its activity. Pemetrexed also has anti-inflammatory properties and can be used to treat inflammatory bowel disease. This drug is metabolized by esterases into glutamic acid, an acid that can lead to kidney stones and other side effects. The hydroxyl group on pemetrexed reacts with acidic molecules such as ascorbic acid (vitamin C) or uric acid (a product of metabolism), forming a salt which can be excreted from the body in urine.
