Glycine 7-amido-4-methylcoumarin hydrochloride is a fluorescent probe that is used to measure the activity of proteasomes. It is a substrate for enzyme activity and can be hydrolyzed by enzymes, which leads to fluorescence emission. Glycine 7-amido-4-methylcoumarin hydrochloride has been shown to inhibit the proteasome in vitro and in vivo, leading to an accumulation of ubiquitinated proteins. This inhibitor also inhibits debranching reactions, which are important for muscle protein synthesis. The enzyme activities of glycylglycine 7-amido-4-methylcoumarin hydrochloride have been shown to be present in all cell types and are not inhibited by the presence of ATP or Mg2+.
