Description

3-Hydroxymethyl-s-triazolo[3,4-a]phthalazine (3HMTPA) is a hydralazine metabolite that is formed in the liver through the acetylation of hydralazine. It has been shown to be an effective drug for the treatment of hypertension, with a dose-dependent increase in blood pressure and heart rate. 3HMTPA is also metabolized to phthalazinone, which has been shown to have cardiotoxic effects at higher doses. 3HMTPA has been found to have bimodal activity with both acetylators and non-acetylators. Acetylators are those who can convert hydralazine into 3HMTPA and other metabolites; non-acetylators cannot do this conversion. Non-acetylator patients may require lower doses than acetylator patients because they cannot produce as much 3HMTPA as acetylators can.
