Tosyl-L-proline (TP) is a ligand that binds to the active site of the enzyme salicylhydroxamic acid (SAHA) and inhibits its activity. TP has been shown to be a potent inhibitor of SAHA in vivo, with greater than 50% inhibition at concentrations as low as 10 μM. TP is also an effective inhibitor of piperazine adenine dinucleotide phosphate (PAP) reductase, which catalyzes the conversion of piperazine adenine dinucleotide phosphate into nicotinamide adenine dinucleotide phosphate. The TP ligand binds in a different orientation than SAHA, which may account for its higher potency. The TP molecule is chiral and can exist in two orientations; one where the chlorine atom is on the left side, and one where it is on the right side. This difference in orientation leads to differences in activity between the two versions of TP.
