Olcegepant hydrochloride is a potent and selective inhibitor of the calcitonin gene-related peptide receptor (CGRP) that is involved in the transmission of pain signals. Olcegepant hydrochloride binds to CGRP with high affinity, but not to other related receptors, such as the adrenergic alpha-1 receptor or the vasopressin receptor. The binding of olcegepant hydrochloride to CGRP inhibits neuronal activity, which may be due to its ability to inhibit ion channels and ligand-gated ion channels. Olcegepant hydrochloride has been shown to block the activation of nociceptive neurons by CGRP in animal models and may be used for the treatment of migraine headaches and other pain disorders caused by overactive CGRP receptors.
