2-(4-Fluorophenyl)-1-tosylpyrrolidine (FPP) is a potent and selective allosteric modulator of the dopamine D4 receptor. FPP is a competitive antagonist at the D4 receptor, with a Ki value of 0.3 nM. FPP acts as an allosteric modulator of the D4 receptor, potentiating the effects of dopamine by binding to a site that is distinct from the orthosteric binding site. This compound has been shown to reduce depression in animals and may have some utility in treating depression in humans. FPP has also been shown to modulate glutamate, however its mechanism of action is not yet clear.
