3-Iodo-5-nitro-1H-indazole is an intermediate in the preparation of biologically active indazoles. It can be synthesized by a cross-coupling reaction with boronic acid pinacol ester as a starting material, followed by selective iodination. The methodology has been applied to the synthesis of 3-(3-methoxypropyl)indazole, 3-(4′-methylpentyl)indazole, and 3-(4′-methylhexyl)indazole. The method has also been applied to the synthesis of 2-(vinyloxymethyl)indazole, which is an intermediate for the synthesis of 2-[2-(2,6-dichloroquinolin-4-yloxy)]ethyl]indazole. This synthetic process provides yields that are more than 95%.
