Podophyllotoxin ist ein natürlich vorkommendes Lignan, das aus den Wurzeln der Mayapple-Pflanze, *Podophyllum peltatum*, gewonnen wird. Es ist durch seine komplexe polycyclische Struktur gekennzeichnet, die ein Dibenzylbutyrolacton-Skelett umfasst. Diese Verbindung zeigt eine signifikante biologische Aktivität, insbesondere als Antitumoragent, und wurde zur Behandlung verschiedener Krebsarten eingesetzt, einschließlich Hodenkrebs und bestimmter Arten von Hautläsionen. Podophyllotoxin wirkt, indem es das Enzym Topoisomerase II hemmt, das für die DNA-Replikation und Zellteilung entscheidend ist, was zu einem Zelltod in schnell teilenden Krebszellen führt. Darüber hinaus besitzt es antivirale Eigenschaften, insbesondere gegen das humane Papillomavirus (HPV), was es nützlich für topische Behandlungen von Warzen macht.
