• Descripción: Simvastatin is a hydrated, acylated statin that is used as a lipid-lowering drug. It inhibits HMG-CoA reductase, an enzyme that plays a central role in cholesterol biosynthesis. The reaction solution can be activated by mixing with an acyltransferase and the desired acyl group. Simvastatin has a toxicity profile that includes myopathy, rhabdomyolysis, and liver failure. Simvastatin also inhibits lipoprotein lipase and prevents the hydrolysis of triglycerides to fatty acids and glycerol. This leads to reduced levels of low-density lipoproteins (LDLs) and increased levels of high-density lipoproteins (HDLs). Simvastatin is converted into an amide form by enzymatic cleavage, which may lead to metabolic inhibition.
