Sim-9 is a covalent allosteric inhibitor of the interferon regulatory factor 3 (IRF3). It covalently binds to the Cys222 residue of IRF3, causing a conformational change that blocks its interactions with TRIF, MAVS, and STING, thereby inhibiting IRF3 homodimerization and the type I interferon response. In mouse models of sepsis and acute pancreatitis, Sim-9 demonstrates significant anti-inflammatory, organ-protective, and survival benefits. This compound is applicable for research into inflammatory diseases.
