It binds to the extracellular domain of the alpha subunit and blocks voltage-gated sodium channels, which are necessary for action potential propagation. GNE-0439 is a potent inhibitor of neuronal and cardiomyocyte Na(+) current, with an IC50of 2 nM in rat neurons and 1 μM in human atrial myocytes. In addition, GNE-0439 inhibits voltage-gated potassium channels (IC50 = 200 nM) and glutamate receptors (IC50 = 2 μM).
