2-(1H-Imidazol-1-yl)benzonitrile is a potent inhibitor of cytochrome P450 enzymes. It inhibits the hydration reaction in the monooxygenase system, which is necessary for the conversion of substrates to products. 2-(1H-Imidazol-1-yl)benzonitrile has been shown to cause centrilobular necrosis in rats and intensifies chlorobenzene toxicity by inhibiting hepatic glutathione synthesis. This compound also causes hepatic microsomal enzyme induction and increased levels of cytochrome P450 in rat liver microsomes, resulting in an increase in hepatocyte metabolism. 2-(1H-Imidazol-1-yl)benzonitrile also inhibits epoxide hydrolase, leading to increases in epoxides such as 3-methylcholanthrene and styrene metabolites, which are both carcinogenic compounds.
