Quisinostat is a potent and selective inhibitor of histone deacetylase (HDAC) enzymes. HDACs are a family of enzymes that regulate gene expression by removing acetyl groups from acetylated lysine residues on histones and other proteins. Quisinostat is biodegradable, radiosensitizing, and has shown efficacy in animal models of cancer. It disrupts the HAT-dependent transcriptional repression complex in the nucleus to promote expression of genes involved in cellular proliferation. It is also an inhibitor of catalytic subunit of DNA polymerase δ, which is required for replication at the site of DNA damage induced by radiation or chemotherapeutic agents.
