2-Imino-2H-chromene-3-carboxamide is a synthetic compound and a potent inhibitor of bacterial DNA gyrase. The functional theory for the inhibition of 2-Imino-2H-chromene-3-carboxamide was first proposed by D.L. Corey in 1973. The active methylene group interacts with the enzyme's metal ion to form a covalent bond, which is thermodynamically unfavorable. This interaction leads to significant cytotoxic effects, as well as inhibitory activities against GyrA, GyrB, and TopA enzymes. Tautomeric equilibrium between the two possible tautomers of 2-Imino-2H-chromene-3-carboxamide has been shown to be important for its inhibitory activity on DNA gyrase.
