Spiperone is a drug that binds to dopamine receptors and has shown high potency and affinity. The 1-Biphenyl-4-yl-piperazine structure is hydrophobic, which may be due to the pyridine substitutions on the aromatic ring. Spiperone has been shown to block the exchange of guanosine triphosphate (GTP) for guanosine diphosphate (GDP) in the G protein system, thus inhibiting the activation of adenylate cyclase. This inhibits the synthesis of cyclic AMP (cAMP), leading to a decrease in intracellular levels of cAMP. Spiperone has also been shown to bind to dopamine receptors and has been used as an antihypertensive agent.
