2-Chloro-4-methoxynicotinonitrile (CLMN) is a drug that inhibits the IKKβ kinase. It is a novel and selective inhibitor of the IKKβ kinase, which is found in the NF-κB signaling pathway. CLMN inhibits inflammation by blocking the activation of NF-κB and inhibiting its downstream gene transcription. CLMN has been shown to inhibit LPS-induced TNFα production in human monocytes. This drug has also been shown to be effective against other inflammatory cell types, such as macrophages, neutrophils, and dendritic cells. CLMN has potential as an anti-inflammatory agent for inflammatory diseases such as asthma and arthritis.
