8-Chloroquinazoline is a glutamic acid substituted quinazoline that has been shown to have potent inhibitory activity against the amyloid precursor protein (APP) in vitro and in vivo. It has also been shown to be an electron microscopic marker for Alzheimer's disease. 8-Chloroquinazoline can be used as a chemotherapeutic agent for Alzheimer's disease, but it is not yet known whether it is effective in humans. The compound is structurally related to curcumin and donepezil, which are vitamin B6 derivatives that are used for the treatment of Alzheimer's disease. 8-Chloroquinazoline inhibits APP cleavage by binding to the active site of β-secretase and γ-secretase enzymes, preventing the formation of β-amyloid peptides.
