3,4-Dihydro-1H-1,4-benzodiazepine-2,5-dione (3,4-DHBD) is a tertiary butyl analog of the diazepinone nucleus. It has been shown to be a potent ATP analog with high specificity for the bace1 enzyme. The compound has also been shown to have anticancer activity in vitro and in vivo by inhibiting cell proliferation and inducing apoptosis. 3,4-DHBD binds to the N-terminal domain of bace1 with high affinity and selectivity; it does not bind to other proteins such as cyclophilin A or alpha synuclein. 3,4-DHBD is also an effective inhibitor of protein synthesis and is in preclinical stage of development for treatment of Alzheimer's disease.
