2-(2-Ethoxyphenyl)-5-methyl-7-propyl-3H-imidazo[5,1-f][1,2,4]triazin-4-one (MIA) is a competitive inhibitor of phosphodiesterase type 5 (PDE5). The recovery and validation of MIA was performed by analyzing the drug in human serum with an electrospray ionization mass spectrometer. MIA was found to be stable in human serum. MIA was quantified and its concentration was determined to be within the range of 0.02±0.04 μg/mL. This study also validated an analytical method for quantification of MIA in human serum using reversed phase HPLC with UV detection at 450 nm and reaction monitoring at m/z 214.
