Epigriseofulvin is a naturally occurring antibiotic that has been isolated from Penicillium griseofulvum. Epigriseofulvin displays high activity against dermatophytes and other fungi, with its most potent activity being against Microsporum canis. It inhibits the synthesis of ergosterol, an essential component of fungal cell membranes, by binding to the enzyme 14-alpha-demethylase. Epigriseofulvin also has low toxicity for humans, as it does not cross the blood-brain barrier and is readily excreted. The chemical modification of epigriseofulvin leads to its increased potency against dermatophytes as well as microbial transformation into a more active form in vivo.
