6-Ethyl-5-(4-fluoro-3-nitrophenyl)pyrimidine-2,4-diamine is an antifolate that inhibits the enzyme dihydrofolate reductase. The ligand binds to the target enzyme with a high affinity and the binding affinities of the two isomers are different. The enantiomeric mixture of 6-ethyl-5-(4-fluoro-3-nitrophenyl)pyrimidine-2,4-diamine has been shown to have potent antifolate activity against methotrexate resistant cells in vitro and in vivo. Spectroscopy studies have shown that this molecule exists as enantiomers, which can be interconverted by reductases (e.g., NADPH).
