4-fluoro-1H-indole-7-carbonitrile is a prodrug that is converted to the active form 4-fluoro-[1,2,4]triazolopyrimidine in the liver. It has potent inhibitory activity against HIV-1 protease and has been shown to be an effective inhibitor of tetrazole. The oral bioavailability of 4-fluoro-[1,2,4]triazolopyrimidine was enhanced by coadministration with acidic compounds such as citric acid. The plasma concentration of 4-fluoro-[1,2,4]triazolopyrimidine was increased by about 10 times when taken orally as a prodrug. This drug has been shown to have a low oral bioavailability (about 10%).
