2-Fluoro-3-methoxybenzoic acid (2FMBA) is an intramolecular lithiated compound that has been shown to be a potent inhibitor of the enzyme Dihydrofolate reductase. It is synthesized by reacting lithiated fluorobenzenes with methoxide ions in an aqueous solution at pH 10. The synthesis yields are improved by using lithium diisopropylamide as the base and 2FMBA as the product. Experiments have shown that 2FMBA inhibits bacterial growth, binds to DNA, and inhibits RNA synthesis in bacteria.
