cis-2-Fluorocyclopropaneamine is a synthetic, non-steroidal antimicrobial agent. It is a potent inhibitor of bacterial topoisomerase IV, which maintains the integrity of bacterial DNA. Cis-2-Fluorocyclopropaneamine also has activity against Gram-positive bacteria and fungi. The synthesis begins from benzyl chloride, a Grignard reagent, and chloroformate as a chlorinating agent. This compound can be prepared by reaction of vinyl acetate with tosylate or p-toluenesulfonic acid in the presence of 2 equivalents of quinolinecarboxylic acid. Cis-2-Fluorocyclopropaneamine can also be prepared by a two step sequence that involves the hydrolysis of 3-(4'-fluoroacetophenoxy)propionic acid followed by the addition of ethyl magnesium bromide.
