Descrizione

Cyclosporin A is a cyclic peptide that binds to the outer mitochondrial membrane and blocks calcium ion (Ca2+) channels, which inhibits the release of Ca2+ from mitochondria. This leads to an increase in free cytosolic Ca2+, which can activate protein kinase C and phospholipase A 2, resulting in increased lipid synthesis. The drug has been shown to block the opening of K channels and inhibit the release of Ca2+ through voltage-gated channels, which causes hyperpolarization of the cell membrane. These effects are due to its ability to bind with high affinity to fatty acids, such as oleic acid, found on the inner surface of membranes. Cyclosporin A also increases cyclic adenosine monophosphate (cAMP) levels by inhibiting cAMP phosphodiesterase activity.
