L-Homohistidine is a homologue of histidine that has been identified as a potential mechanism for the treatment of pancreatic cancer. L-homohistidine is synthesized from histidine and has an inhibitory potency in the range of 0.1-1 μM against polypeptides such as insulin and amylin, which are important for the function of pancreatic cells. L-Homohistidine also inhibits tumour growth by preventing cell proliferation and blocking glucose uptake. This drug may be useful in inhibiting the production of reactive oxygen species (ROS) by inhibiting enzymes involved in ROS synthesis, such as oxidases and oxide synthases. L-homohistidine also binds to imidazoles and forms covalent bonds with them, which can lead to conformational changes in the protein's active site, thus altering its function.
