Duloxetine is a selective serotonin and norepinephrine reuptake inhibitor (SNRI) that is used to treat major depressive disorder, generalized anxiety disorder, and diabetic peripheral neuropathic pain. Duloxetine is a racemic mixture of the two enantiomers, (+)-duloxetine and (-)-duloxetine. The pharmacokinetic properties of each enantiomer are not identical because of their different routes of metabolism. Duloxetine interacts with human cytochrome P-450 enzymes in the liver to produce its active metabolites, which are responsible for the drug's therapeutic effects.
Duloxetine has been shown to be effective in the treatment of depression and anxiety disorders by preventing serotonin from being reabsorbed into the presynaptic neuron, thereby increasing its availability in the synaptic cleft.
