Maduramicin is a quillaja saponaria-derived natural product that exhibits potent antibacterial activity with a novel mechanism of action. It inhibits the synthesis of bacterial DNA and RNA, which leads to cell death by inhibiting protein synthesis. Maduramicin has been shown to be active against drug-resistant Staphylococcus aureus and Mycobacterium tuberculosis. Maduramicin also induces caspase-independent cell death in human cells, but not in murine cells. This effect is due to its ability to inhibit mitochondrial membrane potential and energy metabolism.
