6-Methyl-1(2H)-isoquinolinethione is a hydrophobic compound that is used as a polymerase inhibitor. It competitively inhibits the incorporation of nucleotides into DNA by binding to the catalytic site of the polymerase and blocking access to the template strand. 6-Methyl-1(2H)-isoquinolinethione has been shown to inhibit replication of HIV, influenza A virus, human cytomegalovirus, and herpes simplex virus type 1 in cell culture. The mechanism of action may be due to its ability to bind tightly to the active site of the enzyme with high sequence specificity, preventing polymerization by forming an inactive ternary complex. 6-Methyl-1(2H)-isoquinolinethione has also been shown to increase rates of nucleotide incorporation in vitro by up to five times when used in conjunction with a surrogate DNA template.
