4-Phenyl-1,2,3,6-tetrahydropyridine (4PHTP) is a pro-apoptotic molecule that inhibits the growth of cells by binding to cytochrome P450 and preventing the synthesis of NADPH. 4PHTP is also an inhibitor of rat striatal and human liver cytochrome P450. It has been shown to inhibit recombinant human cytochrome p450s, with an IC50 value of 0.8 μM. The inhibition is reversible and noncompetitive, as it does not compete with substrate for binding sites on the enzyme. 4PHTP damages DNA in humans, which may be due to its ability to bind covalently to proteins such as histones and proteins involved in DNA repair.
