Rolapitant is a drug that belongs to the class of antiemetics. It has been shown to be effective in preventing nausea and vomiting caused by chemotherapy, as well as postoperative nausea and vomiting. Rolapitant is a P-glycoprotein substrate that inhibits the neurokinin-1 receptor, which is involved in the development of emesis. Rolapitant has been shown to be effective against cancer cells with high levels of cyp3a4 activity, but not against those with low levels of cyp3a4 activity. Rolapitant also binds to the neurokinin-1 receptor on chronic cough patients, thereby reducing their symptoms.
