(S)-(-)-1,2,3,4-Tetrahedro-naphthoic acid is a chiral organic compound that can be synthesized in two different ways. In the first method, it is prepared by reduction of the corresponding nitro compound with sodium borohydride. The second method involves the synthesis of an acetone adduct and treatment with hydrochloric acid. (S)-(-)-1,2,3,4-Tetrahedronaphthoic acid crystallizes from acetone in needles or as a white powder. This substance is used in industrial processes for the production of palonosetron hydrochloride (Aloxi).
A dropwise addition of triphosgene to toluene gives a mixture of diastereomers that can be separated by column chromatography on silica gel. A solution of (S)-(-)-1,2,3,4-Tetrahed
