2,5,6-Trichloro-3-pyridinecarbonitrile (TCPC) is a potent kinase inhibitor that binds to the ATP binding site of the enzyme. TCPC has been shown to have a high affinity for Jak2 kinase and can be used as a lead compound in the development of selective inhibitors for this enzyme. TCPC has also been shown to inhibit other kinases with low selectivity. It is metabolized by cytochrome P450 enzymes and glucuronidases, which are responsible for its pharmacokinetic profile. TCPC can be used in biochemical screening and optimization studies because it does not bind to calf thymus DNA or bacterial DNA gyrase and topoisomerase IV.
