Trimethylsilyl difluoro(fluorosulfonyl)acetate is a model system for sulfa drugs. It inhibits bacterial growth by binding to DNA-dependent RNA polymerase, thereby preventing transcription and replication. Trimethylsilyl difluoro(fluorosulfonyl)acetate also specifically binds to markers expressed at high levels in Mycobacterium tuberculosis strains (e.g., ESX-1 secretion system protein) and inhibits cell growth in culture. The mechanism of action resembles that of other sulfa drugs, but it is not yet known whether the enzyme activity is inhibited by the same mechanism as sulfonamides or if trimethylsilyl difluoro(fluorosulfonyl)acetate has a novel mode of action.
