3-(4-Amino-phenyl)-pyrrolidine-2,5-dione is a potent inhibitor of the enzyme dihydrofolate reductase. This drug binds to the substrate binding site and inhibits the conversion of dihydrofolate to tetrahydrofolate, which is an essential step in the biosynthesis of DNA. 3-(4-Amino-phenyl)-pyrrolidine-2,5-dione has been shown to be active against placental and human cells with IC50 values in the low micromolar range. The enantiomers have different inhibitory potencies and are used as racemates for this drug. Crystallographic studies suggest that 3-(4-amino-phenyl)-pyrrolidine-2,5-dione binds to a hydrophobic pocket in dihydrofolate reductase that contains residues His301 and Tyr304.
