Gefitinib is a small molecule with an unknown mechanism of action. It is a potent inhibitor of the epidermal growth factor receptor (EGFR) tyrosine kinase, which regulates cell proliferation and differentiation. Gefitinib has been shown to bind to EGFRs with high affinity and inhibit the phosphorylation of tyrosine residues in EGFRs, which blocks the downstream signaling pathways that regulate cell growth. This inhibition leads to inhibition of tumor cell proliferation and induction of apoptosis. Gefitinib has also been shown to inhibit ligand-induced activation of EGFR by preventing ligand binding, demonstrating its selectivity for EGFR over other protein targets.
