5-([1,1'-Biphenyl]-4-ylmethyl)-N,N-dimethyl-2H-tetrazole-2-carboxamide is a potent covalent inhibitor of lipases and catalyses the alkylation of the active site. It binds to the catalytic residue of the enzyme, which blocks the nucleophile from attacking the substrate. The drug is used in lipase research as a competitive inhibitor for studying enzyme kinetics. 5-[(1, 1'-biphenyl]-4-ylmethyl)-N,N-dimethyl-2H-tetrazole-2-carboxamide has been shown to inhibit monoacylglycerol lipase (MAGL) and HSL with IC50 values in the low micromolar range. This drug also inhibits preferentially MAGL over other enzymes such as pancreatic lipase (PL).
