10-Aminocamptothecin is a potent antitumor agent that inhibits the activity of DNA topoisomerase II. This enzyme is responsible for the maintenance of DNA integrity and replication. 10-Aminocamptothecin has been shown to be active against solid tumours, including metastatic colorectal cancer, squamous carcinoma, and chronic myelogenous leukemia. It also has low toxicity in humans. The drug can be used as a monotherapy or in combination with other drugs such as radiation therapy or chemotherapy. 10-Aminocamptothecin interacts with CYP3A4 enzyme to produce camptothecin metabolites that bind tightly to DNA and inhibit topoisomerase II activity, leading to cell death by inhibiting the production of proteins vital for cell division.
